DNA replikatsioon on poolkonservatiivne – see tähendab, et replikatsiooni käigus sünteesitakse (kopeeritakse) mõlema vanemahela pealt uus komplementaarne tütarahel. Sünteesi käigus lisatakse desoksüribonukleosiid trifosfaate, lühendatult dATP, dGTP, dCTP ja dTTP. Lisamise käigus lõigatakse ära 2 fosforhappejääki ja DNA ahelasse liitub desoksüribonukleosiid monofosfaat, lühendatult dAMP, dGMP, dCMP, dTMP. Süntees on alati 5’-3’ suunas. Uue nukleotiidi suhkrujäägi 5’ ots liidetakse eelmise nukleotiidi 3’ otsaga fosfaatjäägi kaudu. Samaaegselt toimub DNA lahtikeerdumine ja replikatsioon. Ori-regioonid on alati A-T rikkad, praimeriks DNA või RNA või valguga seondunud nukleotiidid, praimerit on vaja selleks, et polünukleotiidahelal oleks vaba 3-OH ots, kuhu DNA-polümeraas saaks liita nukleotiide. 15
