The principle
limitation of efficiency was found to be the width of injection. Diffusion was only limiting
rith an injection zone length of less than 3 mm, negleting thermal and electrokinetic
contributions.
Hydrodynamic injection (HI)
There are thre main ways to introduce a sample onto the head of the electrophoresis column
via hydrodynamin flow: gravity flow (also known as hydrostatic sampling), application of a
positive pressure at the sample reservoir, and application of a negative pressure (vacuum) at
the detector end. The mere insertion of the capillary into the sample solution can introduce
sample into the capillary and thereby limit separation efficiency. The optimum zone length
